Alex Speers ND, MS Within a single tumor, there are a variety of different cell types. One type, called cancer stem cells, or CSCs, are unique because when they divide, they can produce a variety of different cell types in addition to producing more stem cells. This is important because it means that if CSCs are present within a tumor, that tumor has a potential source for any cell type it needs to sustain itself and grow. Now consider what happens when we try to kill a growing tumor. Even if a treatment successfully kills a significant number of cancer cells, that tumor may still be able to regrow if CSCs remain alive. Think of cancer like a weed, growing wildly inside the body. CSCs are the root, and as any good gardener knows, if you don’t kill the root, there’s a good chance that weed will eventually grow back. buy flagyl 400 Here, we propose a new strategy for the treatment of early cancerous lesions and advanced metastatic disease, via the selective targeting of cancer stem cells (CSCs), a.k.a., tumor-initiating cells (TICs). We searched for a global phenotypic characteristic that was highly conserved among cancer stem cells, across multiple tumor types, to provide a mutation-independent approach to cancer therapy. This would allow us to target cancer stem cells, effectively treating cancer as a single disease of “stemness”, independently of the tumor tissue type. Using this approach, we identified a conserved phenotypic weak point – a strict dependence on mitochondrial biogenesis for the clonal expansion and survival of cancer stem cells. Interestingly, several classes of FDA-approved antibiotics inhibit mitochondrial biogenesis as a known “side-effect”, which could be harnessed instead as a “therapeutic effect”. Based on this analysis, we now show that 4-to-5 different classes of FDA-approved drugs can be used to eradicate cancer stem cells, in 12 different cancer cell lines, across 8 different tumor types (breast, DCIS, ovarian, prostate, lung, pancreatic, melanoma, and glioblastoma (brain)). These five classes of mitochondrially-targeted antibiotics include: the erythromycins, the tetracyclines, the glycylcyclines, an anti-parasitic drug, and chloramphenicol.

Figure 6 Doxycycline inhibits tumor-sphere formation in eight other cell lines. Nov 29, 2017. Tumor heterogeneity can be traced back to a small subset of cancer stem. a potential new treatment of targeting mitochondria dependency.

One of the most commonly held beliefs is that cancer cells contain so many genetic mutations that the only way to deal with them is to destroy or remove them by conventional oncological methods such as surgery, chemotherapy and radiation. New research has shown, however, that by altering one specific oncogene in cancer cells, Myc (pronounced “mick”), one can, in essence, turn “off” certain forms of aggressive malignant cells. Myc plays an essential role in all cellular life cycles. This protein sends the message for the cell to divide. In normal cells, this protein is only made when it’s the proper time for the cell to divide and multiply. In cancer cells, however, Myc is overproduced which results in uncontrolled and constant cell division and replication. This uncontrolled cellular activity results in tumor growth. Background and objectives: Cancer stem cells (CSCs) have been implicated in tumor initiation, recurrence, metastatic spread and poor survival in multiple tumor types, breast cancers included. CSCs selectively overexpress key mitochondrial-related proteins and inhibition of mitochondrial function may represent a new potential approach for the eradication of CSCs. Because mitochondria evolved from bacteria, many classes of FDA-approved antibiotics, including doxycycline, actually target mitochondria. Our clinical pilot study aimed to determine whether short-term pre-operative treatment with oral doxycycline results in reduction of CSCs in early breast cancer patients. Methods: Doxycycline was administered orally for 14 days before surgery for a daily dose of 200 mg. Immuno-histochemical analysis of formalin-fixed paraffin-embedded (FFPE) samples from 15 patients, of which 9 were treated with doxycycline and 6 were controls (no treatment), was performed with known biomarkers of “stemness” (CD44, ALDH1), mitochondria (TOMM20), cell proliferation (Ki67, p27), apoptosis (cleaved caspase-3), and neo-angiogenesis (CD31). For each patient, the analysis was performed both on pre-operative specimens (core-biopsies) and surgical specimens.

In addition to the general indications for all members of the tetracycline antibiotics group, doxycycline is frequently used to treat Lyme disease, chronic prostatitis, sinusitis, pelvic inflammatory disease, Moraxella catarrhalis, Brucella melitensis, Chlamydia pneumoniae, and Mycoplasma pneumoniae are generally susceptible to doxycycline, while some Haemophilus spp., Mycoplasma hominis, and Pseudomonas aeruginosa have developed resistance to varying degrees. Some Gram-positive bacteria have developed resistance to doxycycline. Up to 44% of Streptococcus pyogenes and up to 74% of S. faecalis specimens have developed resistance to the tetracycline group of antibiotics. When bacteriologic testing indicates appropriate susceptibility to the drug, doxycycline may be used to treat these infections caused by Gram-positive bacteria: The World Health Organization Guidelines states that the combination of doxycycline with either artesunate or quinine may be used for the treatment of uncomplicated malaria due to Plasmodium falciparum or following intravenous treatment of severe malaria. Doxycycline kills the symbiotic Wolbachia bacteria in the reproductive tracts of parasitic filarial nematodes, making the nematodes sterile, and thus reducing transmission of diseases such as onchocerciasis and elephantiasis. Doxycycline has been used successfully to treat sexually transmitted, respiratory, and ophthalmic infections. This is my first month on clomid, I took 150mg from days 2-6, and now I an CD 18, at CD 23 I will be able to have an HCG test to see if anything happened. I started off days 5-9, then 3-7 and now I'm trying 2-6. gave me progesterone suppositories to take post-ovulation. My third cycle I did 100mg CD2-6 and we are pregnant. *hugs* I'm your cheerleader around here ;) Thanks guys, im waiting on af to arrive so i can take my first clomid, doc posted me out a prescription for metformin today, but dont know if i should start it now or wait and start it same day as clomid!? I am currently CD 7, Hopefully at the end of this month I should know if I am preggo or not, and will hopefully be a success story. I'm new here and just started my clomid on cd 2, today is cd 4 and I have a question. So Ready, my clomid looked exactly like yours also. I will let you know, and I hope to be a success story, but for now waiting is all I can do! I did end up with two large cysts..on each ovary, but I also got my little bean too :) So, I'm thrilled with the days and dosage I took. I have taken clomid in the past, and with only a 25mg dose -- was having aggressive hot flashes. The first month I got really bad s/e starting day 2 (hotflashes, bad bloating, headaches, cramps, mood swings, nausea, crying for no reason) my second round I didn't have any side effects at all and now the third I have bloating and I have a fuzzy almost dizzy feeling throughout the day. I bought mine in Mexico instead of ordering it online. Ciara~ The other ladies know already so I though I would post for you on your thread :) I took clomid for three cycles. Second dose was 50mg CD3-7 and I conceived twins, but we m/c'd them both. Last month I did 150 mg days 2-6 without success, but AF came on its own after that, so I am hoping that this month, my second round will be the lucky one. It's nice to not have the hot flashes though (knock on wood). My O got pushed to CD 17 with this last round and had we of given up earlier, I would not have a success to speak of ;) BABY DUST to you!!! Just thought you may like to hear my story too :) Good luck and baby dust to you all! This time I'm on Mexican clomid (omifino) it's supposedly the same drug, yet even on 100mg I don't have any side effects. Is it working and I'm just lucky not to be having side effects, or could it be a placebo? Do the side effects happen while your on it, or after I've stopped? The side effects are definately different for everyone. I think it's due to the clomid (I never had this feeling before) So every round I've felt different s/e. If your worried about the authenticity of the medication, you could always take it to your family physician or to your pharmacist so they can check it out. I was a little worried to take it, but my cousin, who is a pharmacist, a__sured me that it was just like clomid. I have just the slightest headache, the kind I used to get on birth control pills, but no hot flashes. My side effects were just a slight nagging headache every day that I was on it, and occasionally a t_tch of nausea. I have slight ovulating cramps now, but I have those every month anyway, and they haven't gotten too severe yet. I think the clomifeno is working though, because since I've posted I've had a dull headache and the slightest bit of a queasy stomache. Nothing else to report as far as my experiences with s/e :) I hope you do not have a darn one of them but are able to tell that you're O'ing ;) HUGE hugs to you and make sure not to give up BD'ing too soon. Linda S - I finally got my clomid in the mail today and I was hoping to compare it to yours. I was a "Just call me Major B" :( Oh how my rage went through the roof..only lasted about 30-45 minutes after taking the pills and then stopped just as quickly as it started, thankfully.
